Atiprimod dihydrochloride is the dihydrochloride salt form of atiprimod, which is an orally bioavailable small molecule belonging to the azaspirane class of cationic amphiphilic agents. It is a JAK2 inhibitor with IC50 value of 397 nM. It has anti-inflammatory, antineoplastic, and anti-angiogenic activities. It inhibits the phosphorylation of signal transducer and activation of transcription 3 (STAT3) and STAT5. It downregulates the anti-apoptotic proteins Bcl-2, Bcl-XL, and Mcl-1 and blocks the signaling pathways of interleukin-6, vascular endothelial growth factor (VEGF). It also inhibits cell growth, induction of cell cycle arrest, and apoptosis in cells expressing the JAK2V617F mutation.
